Blood clot fracture properties are dependent on red blood cell and fibrin content

چکیده
Thrombus fragmentation during endovascular stroke treatment, such as mechanical thrombectomy, leads to downstream emboli, resulting in poor clinical outcomes. Clinical studies suggest that risk is dependent on clot composition. This current study presents the first experimental characterization of composition-dependent fracture properties blood clots, addition development a predictive model for fragmentation. A bespoke test-rig and compact tension specimen fabrication has been developed measure toughness thrombus material. Fracture tests are performed three physiologically relevant compositions: high-fibrin made from 5% haematocrit (H) mixture, medium-fibrin form 20% H low-fibrin 40% mixture. observed significantly increase with increasing fibrin content, i.e. red cell-rich clots more prone tear loading compared fibrin-rich clots. Results also reveal behaviour analogues different compression tension. Finite element cohesive zone modelling experiments show fibres become highly aligned direction perpendicular crack propagation, providing significant toughening mechanism. The results presented this provide key importance next-generation thrombectomy devices strategies. provides characterisation entails novel techniques testing specimens. develops models initiation propagation resistance higher than cell rich Simulations stretching realignment network should be included material order accurately replicate compression-tension asymmetry enhanced toughness. have potentially important implications terms secondary embolization procedures.

Effect of Thyroid Dysfunctions on Blood Cell Count and Red Blood Cell Indice
Abstract Background Thyroid hormones have a crucial role in metabolism and proliferation of blood cells. Thyroid dysfunction induces different effects on blood cells such as anemia, erythrocytosis leukopenia, thrombocytopenia, and in rare cases causes’ pancytopenia. It also alter RBC indices include MCV, MCH, MCHC and RDW. Thus this study attempted to evaluate effect of hypo & hyperthyroi...
متن کاملmeasuring viscoelastic properties of Red Blood Cell using optical tweezers
Efforts have been made to study the behavior of complex materials in micrometer dimensions with various techniques. One of these methods is the use of optical tweezers for biophysical applications. Red blood cells, as the most abundant blood-forming cells, play an important role in the life of living organisms, and their unique mechanical properties are important. In this report, the study of s...
متن کاملAn Overview on: The Red Blood Cell Histogram
The RBC histogram is an integral part of automated hematology analysis and is now routinely available on all automated cell counters. This histogram and other associated complete blood count (CBC) parameters have been found abnormal in various hematological conditions and may provide major clues in the diagnosis and management of significant red cell disorders. In addition, it is frequently use...
متن کاملThe effects of cryopreservation on red blood cell rheologic properties.
BACKGROUND In transfusion medicine, frozen red blood cells (RBCs) are an alternative for liquid-stored RBCs. Little is known about the rheologic properties (i.e., aggregability and deformability) of thawed RBCs. In this study the rheologic properties of high-glycerol frozen RBCs and postthaw stored in saline-adenine-glucose-mannitol medium were compared to those of conventionally liquid-stored ...
متن کاملIncrease in Red Blood Cell-Nitric Oxide Synthase Dependent Nitric Oxide Production during Red Blood Cell Aging in Health and Disease: A Study on Age Dependent Changes of Rheologic and Enzymatic Properties in Red Blood Cells
AIM To investigate RBC-NOS dependent NO signaling during in vivo RBC aging in health and disease. METHOD RBC from fifteen healthy volunteers (HC) and four patients with type 2 diabetes mellitus (DM) were separated in seven subpopulations by Percoll density gradient centrifugation. RESULTS The proportion of old RBC was significantly higher in DM compared to HC. In both groups, in vivo aging ...
